Should the rlog normalised gene expression counts be further normalised?

Hello,

As far as I know the rlog normalisation from DESeq2 inputs raw counts of gene expression. Should the output of rlog be further normalised by the traditional normalisation methods like CPM, TPM, or RPKM, etc before downstream co-expression network analysis?

No, they are already normalized for composition / sequencing depth bias, as is mentioned in the ?rlog help page.
