Hello, I'm currently working with package polyester, and I'm trying to modify the way that the number of reads per transcript is generated (readmat). I actually wanted to use a method inspired on DESeq2 (Materials and methods of the 2014 paper) using the distribution parameters a1, alpha0 and sigmad. It is why I wanted to know if you have a range of values (or the order of magnitude) for each one of these 3 parameters in order to do a 'realistic' simulation.
